I'm avid collector - thought do not sell and really should so there are far more knowledgeable folks to answer this than me.
If you are looking to drop the whole as a lot, look to get about half TCGLow, maybe a little better. 60-75% if you have really strong collection of valuable cards at your local card shop. Same deal for online sales.
If you just want to sell choice singles, ebay is good, just under sell the competition on the cards.
There is no Scrye or Duelist anymore, todays card value is usually based of TCGPlayer.com or StarCityGames.
TCGPlayer has a number of high mid low market prices for cards... if you want to sell a card, look for how much it costs to buy the card to see what you need to sell it at regardless of what the tcglow or tcgmid or tcgmarket value is.
Just because a card is worth $100 bucks, doesn't mean that the cheapest buy option is at $100, could list your card at $100 and never see it sell because everyone is selling at $80

As for how to sell
Sell as lot - easy, fast transaction
Sell select cards - time/energy/effort

My best advice is get a good idea of what you have. Don't just walk into card shop and say whats this worth. I would sort them into groups sub $1 dollar cards, $1-5 cards $5-10. $10-25, $25+
Don't dismiss commons/uncommons, there are uncommons in the $75 that used to be found in $0.10 bins - wasteland for example, revised lightning bolts and dark rituals can be worth a buck etc
Some shops are reputable, others not so much, you could walk in with a beta black lotus and ask whats this worth and they would offer $5 bucks to someone who didn't know it's value.
You also might look at joining some of the magic facebook groups, look for ones with multi thousands of users like MAGIC THE GATHERING buy/sell/trade
