When you are extruding using the control panel, look at the feeder. Is the gear actually rotating? On some people's machines, the gear has become loose. It's held on by a set screw.

How are you testing this? Are you heating up the hotend to temp? When you feed it through by hand are you feeding it until it comes out of the extruder? If not, then you aren't feeding it through....

Try trimming the filament with scissors before inserting into the nozzle, this has helped me a lot of times.
