A DAS has a storage controller built in that allows multiple drives to interface over one connection. Many include advanced features such as RAID and the ability to talk to the drive's controller directly.
The storage controller in an enclosure is far less complicated and only allows one drive to interface over a single connection, essentially acting as a converter. It usually does not allow for transparent interfacing with the drive's controller, which is why you often have to take the drive out to perform firmware updates.
In the rare instance that you do find an enclosure (not a DAS) that supports multiple drives, there is probably a very inefficient hub situation going on.
