Picked up one pair in Bets Buy near by. I have Bose QC35 already, but wanted to try Sony MDR1000. The sound quality of Sony is definitely better then of Bose. But MDR1000 feel less comfortable to me...

Yes. And extremely minute changes to the driver won't be audible. What is "burning in" is your ears adjusting to the sound and accepting them.

Not really. Plenty of audiophile websites recommend breaking in certain models of headphones/earbuds.
I just don't know if this is one of those models.

I tried this set at best buy stores. Sound is not that loud compared to any ear buds, including very cheap ones. Does this need any tuneup? I like the controls and NC.

It doesn't — NC is for constant low-pitched sounds like road noise (passenger in car or bus) or jet engine/wind noise on flights.
