Pi 3 (model b) can be "burned" to boot from usb only (and it can't be reversed), you don't even need a microsd card. For pi 0 and pi 0 w, you have to boot from micro sd, but your root file system...

Certainly possible as there are Pi zero add-on boards (HATs) that add the gamepad stick and buttons. However it's probably cheaper and more practical to use a wireless USB dongle or bluetooth...
