Yes but it wont unwrap MKV files and turn them into streamed MP4's ... at least not that I know of.

Synology has the Plex Media Server, Media Server (Synology's), iTunes Server, Logitech media server, Video Station (Synology's)... I'm guessing that one or all of these will do DLNA?

Well you could remux all of your MKV files into MP4 containers if the video and audio there in are supported. I would just replace the Roku with something that played the native format.
