Just add them and get 32 + 16 unless your board has a max limit of 16. If you have issues then just remove the 16 and sell it on marketplace

Considering DDR4 ram prices are kind of low currently I would probably just grab another 8GB stick unless you have 2x4GB already although some MBs support 4 slots as well.

Ran cpu-z or cpu id and under the memory tab you'll see on the upper right hand if it is running in dual mode or not.
