Removing and remounting the cooler on the CPU isn't totally disastrous especially when done shortly after installing and you are reseating or moving it but you can introduce air gaps and there can be cracks in between the paste. Desktops generally have a lot of pressure on the mounts. But my opinion is that, its a small amount of money for a decent thermal paste, so for a few bucks, I would not risk the chance esp for older and drier paste that has cracks because you will introduce air gaps that can cause some overheating.
