1. Sucralose-6-Acetate is Genotoxic
A landmark study led by researchers at North Carolina State University and UNC Chapel Hill investigated sucralose-6-acetate, a fat-soluble compound formed in the gut during the digestion of sucralose.
DNA Damage: In vitro tests exposing human blood cells to sucralose-6-acetate proved the compound is genotoxic, directly causing DNA strand breaks (clastogenic response).
Pre-existing Contaminant: Trace amounts of sucralose-6-acetate are already present in off-the-shelf commercial sucralose before you even ingest it.
Exceeding Safety Thresholds: The European Food Safety Authority (EFSA) sets a toxicological threshold of 0.15 micrograms per day for genotoxic substances. Researchers found that the trace amount of sucralose-6-acetate in a single daily sucralose-sweetened drink can exceed that safety limit, even before accounting for the additional amount your gut produces as it metabolizes the sweetener.
2. Both Sucralose & its Byproduct Cause "Leaky Gut"
When researchers exposed human intestinal epithelial tissue (the cells lining your gut wall) to both sucralose and sucralose-6-acetate, both compounds impaired gut barrier function:
They degrade the tight junctions, the cellular interfaces that seal your intestinal lining.
This increase in epithelial permeability is the literal definition of leaky gut, allowing undigested food particles, toxins, and waste to cross into the bloodstream instead of being excreted. This can wreak havok all over the body, including the brain.
3. Elevated Inflammation and Cancer-Related Gene Activity
The research didn't stop at physical barrier breakdown. When gut tissue was exposed to sucralose-6-acetate, gene expression analysis showed a significant spike in genes associated with inflammation, oxidative stress, and carcinogenicity.
Claiming sucralose is completely harmless unless consumed by the truckload completely glosses over how its chemical metabolites damage cellular DNA and compromise the intestinal lining at realistic daily intake levels.
