Converted the drive to 512e and the drive properly showed up in ESX 6.7 as a drive instead of an unknown memory controller. Previously I had to use hardware passthrough to be able to use it.

This won't work. Having the partitions aligned doesn't affect the way that the data is referenced in regards to 512 sector or 4k. This is at the hardware level.
