http://calibre-ebook.com/ That should help you either get it on there or convert it to a format you can use. I assume you are trying to use the kindle e-reader right? Can't you just download a PDF...

Have a 450mb pdf file that I want to transfer to my kindle fire. I tried storing it on cloud storage on amazon but it doesn't show up on the kindle is there any other way I can transfer the file?

With a PDF that large you might want to break it up first. I would think as one piece performance loading it would be terrible.
