You can mount a dollar AC filter to a box fan. The hepa filters will reduce airflow. For your sq. footage your need multiple box fans or a whole HVAC system.

The square footage that you want to cover is a lot for a home air purifier to handle. You might want to look into installed units, vs. the regular home air purifiers. You didn't mention a price...
