I usually find that defensive decks get me farthest with defect. If you have a way to increase focus, put a lot of frost orbs in play and increase orb slots, then you're set. Using this as a base, there's lots of scaling strategies that will generate the damage output. The great thing about this strategy is that you're mostly looking for uncommon and common cards, which makes it a bit more consistent.
EDIT: After posting this I played ~10 games to beat A20 on Defect. A20 is unfairly difficult, which is why I find A10 to be a lot more fun. My winning run was a Snecko Eye run. Snecko Eye plus cards with high base cost (meteor strike FTW) is really powerful. I got the mummified hand, which synergizes especially because Defect is a power heavy character.

Otherwise there is the turtle build which goes heavy on frost orbs but it can be kind of a slow start unless you have the right relics and cards. But if you have the right stuff it's probably the strongest build.
Other than that, great game. Goes on sale for PC on Steam/GMG from time to time between $8-10.

It's pretty much the ONLY game I play on my Switch undocked, because the Switch's handheld ergonomics make more action/platformy-type games hurt my hands.
