I already bought and received my s22+ but didn't do the tmobile activation since it was shipped to me, if I go in store will they allow me to return and buy again with the same promotions or price etc but this time do the tmobile activation for the promo?

I don't have a t mobile number and what would be the quickest way to get the $250 t mobile bonus? Also what is a line upgrade? Is it a port in or upgrade the phone plan from like $15/mo to $30/mo?

The only way to get that additional discount is to have a TMO account. Line upgrade is when you go to BB and buy a new phone to replace your current phone. No need to port in or change your plan.
