Yes it will work without a problem, the main router can take full advantage of your 500Mb fiber but the nodes will not they will be limited to anywhere between 200-300Mb to a single device but that should be plenty for most people. If you hardwire them all to each other it will increase the speeds on the nodes. If you leave them purely wireless expect the above speeds.
Your optimal setup is:
A - if you can connect units to each other via wires, dual band is totally fine and great (especially due to price point)
B - if you need the satellite units to connect wirelessly (because you cant connect them to each other with a wire), then tri-band is your ideal system so they can dedicate the extra band to wirelessly transmit to each other

From ASUS's website [asus.com], I see that the 3-Pack can cover 4800 Sq. Ft. and 5+ Rooms and the product title states "home coverage up to 4800 Sq. Ft."

In short only matters to people with 1 gig plans, and only really matters if you really want full speeds, if you just care for reliable coverage this will work great and for most devices even 200-300Mbps is more than plenty for anything and everything.
If you are able to wire the nodes to each other you'll get full speeds.

Right now, I think the Asus XT12 are phenomenal for a wireless setup with triband. I like the Asus control suite as well. 2 units, but very powerful coverage wise. It also performs really well for 2.4ghz, which I have a ton of devices using too (security cams, lights, pet feeders, etc all still use 2.4ghz usually). But again, my personal opinion.
