In a pinch, you can get by without an adapter for a little while, but the larger hole in your rim will likely lead to a leak near the valve stem.
Back in the day, we used lamp cord grommets from the hardware store, but they now make nicer aluminum [evergreenmountainsports.com] and rubber [rivercitybicycles.com] grommets for this exact purpose.

A rim drilled for Shrader has a wider opening than a Presta drilled rim. So if you tried to use a presta tube with no modification, the tube could get pinched in the extra width of the hole and puncture. There are some valve adapters (rubber grommets) you can use to make this work, but not worth the trouble vs. just sticking to the shrader tubes.
Assuming you mean current tubes (not tires). Yes this will sub in fine if your existing valves are Presta.
