I built/sold a bunch of aircooled gaming machines in these in 2020. When done up right (like with an IcyDock SSD chassis in one of the bays) they make a great easily-expandable system for nontechnical gamers.

Plexiglass won't have the clarity of tempered glass and won't be as scratch resistant but it's considerable cheaper, easier to DIY, and sturdier. A lot of aquariums are made of it, many of which are part of DIY builds.

If it's a piece of glass inside a larger side panel, the easiest option is to measure the size of the hole and order a new piece of custom-cut glass (or acrylic) from Ebay. Some hardware stores will cut a $10-20 acrylic garage door window panel down a couple of inches, although not many these days. Ace Hardware is your best bet.
If it's a whole-side panel with holes drilled in the glass to attach to the case, it's usually better to get a new case. Maybe you can find a replacement panel on Ebay from someone parting out the same model you have, but drilling glass or acrylic on one-off jobs will be too expensive for a custom replacement.
